I have just installed a new carb on my 641. After ensuring the ignition was in proper working order, the engine will not start. Sediment bowl is full and gas will drain from bottom of carb indicating gas in bowl. There is plenty of vacuum and the carb is secure to intake. The tractor will run when starting fluid is sprayed into intake. At wits end.

My guess is that new carb has blocked main and idle jets and venturi..

Hold your hand over the carb intake and crank it. You should feel a strong vacuum on your hand and the engine should fire quickly. You leave your hand there long and it will flood gas out the intake and then you will know gas is getting through of the carb.
If it can"t get it to fire that way then you have to take a hard look at the spark. Do you get a regular spark at the plug? Measure the plug gap and the points gap. Look closely at the condition of the points. If they are burnt, worn or missing then replace them. Check the timing. Often times, it"s not enough that there is some spark at the plug. The spark has to be strong enough and arrive at the right time.

I have cranked the engine with hand over intake and with choke and no gas flood out the intake. There is significant suction as evidenced by the ring on my hand. There is gas in bottom of the carb when I loosen the plug in bottom of the carb. Points, plugs and spark all check well. As I mentioned, when I spray starting fluuid in the intake and crank it will run. I have also opened up the main jet beyond the 1 1/4 turns with no change.

You say it "runs" when you use starting fluid. Does that mean it "runs" so you can drive it around the yard or do you mean it "fires" on starting fluid but dies when you stop the either?

If it "fires" on either and then starts and "runs" around the yard you should pay attention to the ignition but that doesn"t go with your mention you get no gas out of the carb. I"m guessing when you say "run" you mean "fires."

If no gas is getting through the carb then the tractor isn"t going to "run" around the yard and you must then remove the carb, disassemble the carb and clean it up or repair as needed. You can figure that because it will "fire" (spark) on either but you are not getting any gas out the carb. You may or may not need a carb kit but most guys can get off $25 to get it done while they have it apart to clean. It"s a good idea. If the carb needs repairs you might need a more expensive kit or even piece parts.
It"s not your immediate problem but pay close attention to get all, even the very tiny, ports and passages clean. Check by dabbing some solvent in one end of each passage and see that the solvent comes out the port(s) on the other end. You might find a round, wooden tooth pick and a set of welder"s tip cleaners handy to clean up the carb. Paint thinner (aka mineral spirits, stoddard solvent, white spirits, dry cleaning fluid) is a fair solvent, easy to find, relatively safe to handle, not expensive, and a lot of us generally have some around.
