Swapping the plug and wire to another cylinder and the problem doesn't move will eliminate the wire and plug. Ounce the wire and plug are eliminated, do a good spark test on that cylinder that is missing. A cracked distributor cap will sometimes kill the spark to 1 or more cylinders. If you are getting good spark then that will most likely eliminate problem with cap. Then if problem isn't found remove valve cover and check for stuck valve. If no stuck valve then do a compression chek. The valve or seat may well be burnt.
