First find out which cylinder it is. Pull off one plug wire at a time (using plug wire pliers or well insulated pliers) as each is removed and replaced, one will make no difference. That is the focus. Change the plug in that cylinder with another and retest to see if it followed the plug, if so replace the plug, if not, pull the plug from the cylinder that is not working and test the compression again (with throttle open) If good compression, we need to focus on intake. Use the choke to richen up the mixture till it begins to run rough (not full choke) and see if at some point the engine picks up that cylinder. If so there may be a vacuum leak letting air in that runner of the manifold. Use an unlit propane torch (outside) while it is running and missing. Pass the torch nozzle around the intake manifold to see if it picks up the cylinder. If so there is your leak. If not, I would pull the valve cover off and look at the valve train to see if a valve is stuck, or not moving. Worn out cams, misadjusted valves, and broken bent pushrods will cause that miss.
