In the Ford line at least, the F-150 line and the Super Duty(F-250 through F-550) lines are two completely different trucks. Generally speaking, the bigger trucks will have heavier frames with more stiffening crossmembers, stiffer shocks/springs, heavier axles, bigger brakes, etc. The heavier trucks will also get you bigger engines(including diesels, V10s, etc.) and depending on the engine selection, beefier transmissions. Ford will start offering a new 4.5-liter Powerstroke V6 turbodiesel in the F-150 in 2004.
