Flat spot in tire

I purchased a tractor that had sat for at least 12 years. The rear tires are in very good condition except that one of them has a flat spot from sitting so long. When you drive the tractor it feels like you drive into a hole every 15 feet. At high speeds the whole thing shakes. Will this flat spot ever come out? Is there anything I can do to get rid of it? I hoped that if I drove it around a while it would go away. It has not happened so far. My next idea was to over inflat the tire. Like 18 psi for a while. The tire is a 10 ply. Any suggestions?
