It would help a bit to know your tastes and budget. There Is just a huge variety of places to go in this country.
If you don't mind cities, I highly recommend Washington, DC. You could spend two weeks there and not see everything there is to see. You could easily spend a week in the various Smithsonian museums alone. If you go there in the summer, try to see the Friday night parade at the Marine Corps Barracks, it is one of my favorite memories of DC. (You need to get there early if you don't have reservations.)
If you're looking for something less urban, consider a trip through California. You can visit the beach, the mountains and the desert in one trip. And even though LA and San Francisco are both insane, there is a lot to see and do in either city. The wine country of Napa and Santa Barbara is also popular. The biggest problem with a trip to CA is deciding what you're going to miss. The sheer size of the state dictates that you can only see a small fraction of it in one trip.
I love the North Carolina Outer Banks, but they're not everybody's cup of tea. They are quite remote, and taking a side trip means a long drive down a two-lane highway or possibly a ferry. If you go in the summer, you'll want to try to reserve a cottage; in the fall and winter the beaches will be deserted and you'll have them to yourselves.
Everyone should see Yellowstone before they die. 'nuff said.
The Colorado Rockies are a great trip, there are many spectacular drives you can take.
My wife and I took and Alaska cruise last year. We had a great time and plan to return to Alaska in the future.
