Could be many, many things, and as Allen already stated, the fix is rarely simply. Otherwise, seems the seller would of already taken care of the problem.
If it runs well otherwise, doesn't smoke, and is just missing on one cylinder - could be a bad injector, a bad delivery valve (if a CAV rotary pump), a stuck injector-pump plunger (if an inline pump), a bad valve/seat/rocker arm, etc.
If the whole engine skips and sputters when you rev it up, but smooths out under a load - the timing advance is probably sluggish. Easy fix on some machines, and not so easy on others.
If the engine starts hard and needs ether, skips and smokes until you get it good and hot, the top piston rings are probably broken, as well as several other parts.
