Vapor lock is when gasoline vaporizes (boils) in the fuel line ahead of the carburetor and interrupts the flow of fuel to the carb. It's caused by high ambient temperatures combined with high vapor pressure fuel (i.e. "winter" gas used in hot weather). A fuel tank placed below the engine and requiring a pump to suck the fuel from the tank can also develop vapor lock due to the low pressure on the suction side of the pump causing the gas to boil in the line ahead of the pump. And as Dan mentions below a plugged tank vent can cause vapor lock when the pressure in the tank drops low enough for the gas to boil.
