The thermosiphon depends on the difference in density between hot and cool coolant. If you don't mix your antifreeze and water, you are introducing another density gradation that is even greater than the thermal density gradient. If the antifreeze and water don't mix sufficiently, that density gradation will inhibit circulation. I don't know if you can ruin an engine that way, as the mix will start circulating (or erupting) when the water starts to boil, but probably not worth the risk.
